Lamar is always good for wildlife, regardless of the season. Exactly what you'll see is largely contingent upon what the weather has been like (early cold will push more critters down into the valley, more critters in the valley will bring the wolves down, etc.). Check locally when you get to the park to learn what people are seeing and where.

You shouldn't have an issue with roads (unless there's some project slipping my mind here...check the road construction page just in case). I wouldn't let the weather be a concern. Mid-September roads will be dry unless a rain system is moving through. While there's a chance of seeing flakes up high, mid-Sept. would be early for anything horrific. I've driven the routes you'll be on into late October the last few years and never had an issue with weather conditions other than fog. I wouldn't let this trouble or concern you in the slightest.

I can't speak to specific places to stay in Silver Gate and Cooke City, but your options would range from the Super 8 to various cabins and lodges. I'm not sure when Montana's hunting seasons start, but if they're open on that weekend, that will definitely impact availability for rooms/cabins. Sorry I can't help you more there, hopefully someone else on the page can.
